[atbench]Nanopore Sequeincing, SQK-LSK109に用いるNEB Quick T4 Ligaseのキットについて
NanoporeのLigation sequence SQK-LSK109ではライブラリ作成の際にNEBのQuick T4 Ligaseを10 uLも消費する。ONTではNEBのE6056 NEBNext Quick Ligation Moduleを指定しているが、実際のプロトコルでは使用するのはQuick T4 Ligaseだけであり、バッファーは使わない。またこのQuick T4 Ligaseが入ったNEBのキットは複数あり、M2200 Quick Ligation Kitがセールされている場合は割安であることに気づいた。
気をつけなければいけないのは各キットで使用可能回数を書いているが、下記のURLの通りプロトコルが違うので実際の価格はそれほど違わない点である。

結局、今回はM2200Lを購入した！
Nanoporeのコミュニティにもコメントする形で以下のように記しておいた。
I just found M2200S and M2200L are in sale now (2020/07/17) and so compared the cost performance. In Japan E6056S and E5056L (not T) are only available.
According to NEB, T4 ligase itself kits are completely same in these kits, but number of reaction and its standard protocol are different between those. In each standard protocol, T4 ligase is recommend to be used 5 uL and 1 uL in E6056 and M2200.
